From: Chemical inhibitors destabilize HuR binding to the AU-rich element of TNF-α mRNA
Specificity of the inhibitory activity of quercetin and b-40. TNF-α, COX2, IL-6 or cFos ARE RNA was incubated with GST (128 nM), tristetraprolin (128 nM) and HuR (32 nM). After the reaction mixture was treated with quercetin at 0.5 µM (A) or b-40 at 0.25 µM (B) the percentage of RNA complexed with protein was quantified using filter-binding assay. The results are means ± standard deviation (SD) of duplicates from two independent experiments. Statistical analysis was performed by t-test using Graph Pad Prism5 software. A P value of less than 0.05 was considered statistically significant (**P < 0.01, ***P < 0.001).
