Inhibition of the expression of pro-inflammatory mediators by rf3 and rf4 in HaCaT cells. (a) Cells were stimulated with TNF-α and IFN-γ for 24 h and western blot analysis was performed using anti-COX-2 and anti-ICAM-1 antibodies. Data are representative of three independent experiments with similar results, and GAPDH served as a loading control. (b–d) The mRNA levels of pro-inflammatory mediators were determined by qRT–PCR in HaCaT cells stimulated for 4 h with TNF-α and IFN-γ. Data were normalized to GAPDH signal and are presented as -fold change relative to the vehicle-treated group. Results are expressed as the mean±s.e.m. of three independent experiments. #P<0.001 versus vehicle-treated group; **P<0.001 and *P<0.05 versus TNF-α- and IFN-γ-stimulated groups, respectively.
