Phospholipase D (PLD) inhibition increases ionizing radiation (IR)-induced DNA damage. (a) MDA-MB-231 cells were treated with 10 μM PLD inhibitor for 4 h, after which they were exposed to 5 Gy IR. At 48 h, an alkaline comet assay was performed, and the comet tails were observed by fluorescence microscopy. (b) The cells were treated with PLD inhibitor for 4 h and/or exposed to IR. After 1 h, the lysates were analyzed by western blot using the indicated antibody. These blots are representative of the results obtained from three experiments.
