B-cell lymphoma 2 (Bcl-2) silencing by small interfering RNA (siRNA) delays aspirin-induced apoptotic cell death in MCF-7 cells. (a) Bcl-2 protein levels were detected by immunoblotting 72 h after siRNA transfection. (b, c) Cell cycle analysis. MCF-7 cells were transfected with Bcl-2-targeting or scrambled control siRNA and treated with 5 mM aspirin for the indicated incubation periods. The cell cycles were determined by flow cytometry (b) and quantitated by ModFit LT software (c). (d) Cell proliferation was measured 48 h after aspirin treatment using MCF-7 cells transfected with Bcl-2-targeting or scrambled control siRNA. Error bars represent the s.e. calculated from three independent experiments.
