Phosphorylation of B-cell lymphoma 2 (Bcl-2) is associated with nuclear condensation. (a) MCF-7 cells were exposed to 5 mM aspirin for 24 h. Nuclear and cytoplasmic fractions were prepared as described in the MATERIALS AND METHODS and subjected to immunoblotting with the indicated antibodies. Lamin A and α-tubulin were used as nucleus- and cytoplasm-specific controls. C, cytosolic fraction, N, nuclear fraction. (b) MCF-7 cells were treated with 5 mM aspirin and incubated for the indicated time periods, and phosphorylation of Bcl-2 in the nuclear fraction was determined by immunoblotting. (c) MCF-7 cells were exposed to 5 mM aspirin for 48 h. Cells were stained with anti-phospho-Bcl-2 and 4′,6-diamidino-2-phenylindole (DAPI) and then analyzed by fluorescence microscopy.
