Bucillamine scavenges ROS generated by cisplatin in HEI-OC1 cells. (a) To test spontaneous production of intracellular ROS, cells were treated with DCF-DA, a ROS-specific fluorescence dye, for 1 h and then, fluorescence intensity was measured using a flow cytometer. (b) Cells were pretreated with 2 mM bucillamine followed by the addition of 20 μM cisplatin for 24 h. Then, cells were incubated with the dyes, including DCF-DA for hydroxyl radical and 4-amino-5-methylamino-2′,7′-difluorofluorescein for nitric oxides. After 1 h, the fluorescence intensity was measured using a flow cytometer. (c) Relative intensities from FACS analyses are shown in the bar graph.
