Apoptosis is increased in growth hormone receptor (GHR) silenced pancreatic cancer cell lines. (a) Flow cytometric analysis showed that GHR silencing induces apoptosis in PANC1 and HPAC cells. Fluorescence-activated cell sorting (FACS) histogram of cells stained with Annexin V-fluorescein isothiocyanate and propidium iodide. The percentage of early apoptotic (bottom right quadrant), apoptotic (top right quadrant), late apoptotic and necrotic cells (top left quadrant), and live healthy cells (bottom left quadrant) are represented. (b) Percentage apoptosis and cell death in PANC-1 and HPAC cells. (c) GHR inhibition induces apoptosis in PANC1 and HPAC cells. Bax, Bcl-2, caspase 8, caspase 3 and cleaved PARP and β-actin expression was assayed using western blot in GHR silenced PANC-1 and HPAC cells. (d) Densitometry analysis obtained from density ratio between apoptotic markers and β-actin shows significant potentiation of apoptosis in GHR silenced pancreatic cancer cells.
