Impacts of SIRT1-deficient DCs on T-cell proliferation and Th1/Th17 differentiation in co-culture. The CII-pulsed and LPS-stimulated DCs were generated from the immature bone marrow-derived DCs isolated from the mSIRT1 KO (KO) and wild-type (WT) mice. Activated and matured DCs were co-cultured with the T cells from the non-immunized or immunized CIA mice. (a) The T cells’ proliferative response was assessed by the fluorescence of 5(6)-carboxyfluorescein diacetate succinimidyl ester (CFSE) after stimulation with vehicle (VEH) or CII (20 μg ml−1) for 72 h. The dot plots (left panel) represent one of three experiments, and show the proportion (right panel) of CFSE-positive cells. (b) The T cells’ phenotype was determined by flow cytometry.
