Membrane anchoring of scIL-12 in transfected cells. HuH-7, HaP-T1 or HEK-293 cells were transfected with the indicated plasmids. A subset of cells transfected with pC-scIL-12-GPI was treated with phosphatidylinositol phospholipase C (PI-PLC), as indicated. (a) Twenty-four hours later, conditioned media were collected for quantification of scIL-12 by ELISA. Cells were then harvested, stained with an antibody against murine IL-12 and analyzed by flow cytometry. (b) Percentage of cells displaying scIL-12 in their surface.
