The role of H3K27me3-mediated gene silencing during developmental transitions. In pluripotent cells, developmental regulators are repressed and carry bivalent H3K4me3+H3K27me3 modifications. Bivalent marks are reduced in committed progenitor cells in which two categories of H3K27me3-repressed genes exist. Lineage-appropriate genes are activated during terminal differentiation, probably by the active removal of H3K27me3. In contrast, lineage-inappropriate genes are stably repressed by H3K27me3 and other mechanisms, such as different histone modifications and DNA methylation.
