Effect of TET1 knockdown on the mineralization potential of hDPCs. Cells were cultured in GM and DM. (a) ALP activity was measured at day 7. The ALP activity of all of the groups did not significantly differ under normal culture conditions. However, the ALP activity decreased by 54.92% in the shRNA1 group and by 79.52% in the shRNA2 group compared with the control group in the odontogenic differentiation medium. (b) Mineralization was analyzed using alizarin red S staining at day 21. The formation of mineralized nodules was inhibited in the TET1-knockdown hDPCs undergoing odontogenic induction. All of the results represent the mean±standard deviation of three independent experiments (n=3). *Significant difference compared with the control (P<0.05).
