ErbB4-induced cell survival correlates with COX-2 expression. (a) YAMC-ErbB4 cells were exposed to HRG for 15 min with or without the COX-2 inhibitor celecoxib (CCB; 5 μM, 30 min pretreatment). Cell lysates were prepared and ErbB4 phosphorylation was determined by western blot. (b) YAMC-Vec and YAMC-ErbB4 cells were primed with interferon-γ (150 U/ml) overnight, then exposed to TNF+interferon-γ for 24 h with or without CCB. Cell loss was determined by an MTS-based cell count assay. (c, d) LIM 2405 cells were infected with lentiviral particles to express either control or ErbB4 targeting shRNA constructs and selected with 5 μg/ml puromycin. (c) ErbB4 and COX-2 protein levels were determined by western blot analysis.
