(a) The proportion of donor-derived circulating leukocytes that were CD18−/−. (b) The proportion of donor-derived leukocytes in the BM that were CD18−/−. Blood and BM samples were obtained from C57BL/6-CD45.2 hosts reconstituted with a 1:4 or 4:1 mixture of C57BL/6-CD45.1 and CD18−/− fetal liver cells at least 8 weeks after transplant. Donor-derived leukocytes were identified as either WT (CD45.1/CD11a+) or CD18−/− (CD45.2/CD11a−). The percentage of donor-derived neutrophils (Gr-1+), B cells (B220+), and T cells (CD3+) that were derived from CD18−/− stem cells (CD11a−) was determined by flow cytometry. Dashed lines indicate the expected ratio of CD18−/− cells in each population based on the initial proportion of CD18−/− cells in the transplant (80% in the 1:4 group and 20% in the 4:1 group). Data are expressed as mean±s.e.m. (n=6–7).
