Sal B induces disassembly of actin stress fibers. (a) Cells were treated with ML-7 (10−5M), Y-27632 (10−5M) or Sal B (10−5M) for 30 min, followed by incubation in media with or without ET-1 (10−8M) for 30 min. Morphological change of cells was observed by phase-contrast microscopy. Enlarged views of the boxed areas are presented as indicated. Scale bar, 100 μm. (b) F-actin staining. Cells were pretreated with ML-7 (10−5M), Y-27632 (10−5M) or Sal B (10−5M) for 30 min, then stimulated with ET-1 (10−8M) for 30 min. F-actin was stained by FITC-labeled phalloidin. Scale bar, 100 μm.
