Runx2 associated with EMT-related molecules, MMP and vasculogenic factors. (a) RT-PCR. Runx2 silencing results in decreased mRNA expression of SNAI2 and VEGFC in WRO cells. Runx2-silenced 8305C cells show downregulation of SNAI2, SNAI3, TWIST1, MMP2, VEGFA and VEGFC. (b) Real-time PCR quantitatively validates the significant downregulation of genes in both cell lines (P<0.05). (c) Immunocytofluorescence shows decreased expression of Snail2/Slug (SNAI2) and VEGFC in Runx2 siRNA/WRO cells compared with control cells. Downexpression of MMP2 is confirmed in Runx2 siRNA/8305C cells. Cellular nuclei are labeled with DAPI (small square).
