TBK1 inhibition attenuates radiation-induced EMT. (a) A549 cells were transiently transfected with siRNA-specific TBK1 or with control siRNA. After 24 h, cells were irradiated with 8 Gy of 60Co γ-rays and were harvested at 72 h post irradiation. Expression of each protein was evaluated by western blot analysis. β-Tubulin was used as loading control. (b) Immunofluorescence staining for E-cadherin (green) and 4′,6-diamidino-2-phenylindole (DAPI; blue) in control and treated cells at 72 h post irradiation (original magnification × 100). (c) Immunofluorescence staining for vimetin (green) and DAPI (blue) in control and treated cells at 72 h post irradiation (original magnification × 100). (d) A549 cells were transfected with siCtrl or siTBK1 and subjected to a wound-healing assay with radiation or vehicle.
