TBK1 inhibition regulates radiation-induced ZEB1 expression. (a) A549 cells were transfected with a control or a TBK1-specific siRNA. After 24 h post-transfection, cells were left untreated or treated with 8 Gy of 60Co γ-rays, and total RNA were collected at 3 h after irradiation. RT-PCR analysis was done using a primer for ZEB1. The data were reported as means±s.e.m., *P<0.05. (b) A549 cells were transfected with a control or a TBK1-specific siRNA. After 24 h post-transfection, cells were left untreated or treated with 8 Gy of 60Co γ-rays, and protein samples were collected at various time points after irradiation. Western blot analyses were used to detect transcription factor ZEB1. Equal protein loading was verified by the analysis of β-tubulin. (c) A549 cells were transfected with siCtrl, siTBK1, empty vector, plasmid of ZEB1, or were co-transfected with siTBK1 and plasmid of ZEB1. After 24 h, cells were irradiated with or without 8 Gy of 60Co γ-rays for an additional 72 h. EMT-specific biomarkers were detected by western blot analysis. Equal protein loading was verified by the analysis of β-tubulin.
