M2 markers were detected in native, M1, and M2 macrophages. Protein expression of CD204 and CD206 increased after treatment with interleukin-4 (IL-4). Mouse macrophage cell line, RAW 264.7, cells were treated with IL-4 (20 ng/ml) for M2 or 10 ng/ml lipopolysaccharide (LPS) plus 100 IU/ml IFN-γ for M1 for 24 h, then cultured in serum-free Dulbecco’s modified Eagle’s medium (DMEM) for 48 h. After 48 h, the medium was collected for enzyme-linked immunosorbent assay (ELISA) (a), and the cells were collected for western blotting (b).
