TLR4/interleukin-10 (IL-10) signaling has an essential role in neoplastic characteristics of pancreatic cancer cells. In coculture (CC) with M2-polarized tumor-associated macrophages (TAMs), the pancreatic cell lines were treated with either TLR4 siRNA, neutralizing antibodies of TLR4 and IL-10, and lipopolysaccharide (LPS), respectively. After 24 h, proliferation, migration, and proteolytic activities were determined by 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) (a), transwell (b), and zymography (c, d), respectively.
