Effect of MK0626 (M) on apoptosis during tacrolimus (TAC)-induced renal injury. (a and b) In situ TdT-mediated dUTP-biotin nick end labeling (TUNEL) assay and its analysis to detect apoptosis in the renal tissue sections of the experimental groups. MK0626 treatment significantly reduces TUNEL-positive apoptotic bodies compared with TAC. (c–f) Immunoblot analysis of Bcl-2, Bax, Bim, and active caspase-3 in the renal cortex. Note that MK0626 treatment significantly recovers the expression of Bcl-2 and downregulates Bax, Bim, and active caspase-3 compared with TAC.
