Chondrogenic, hypertrophic, angiogenic, and anti-angiogenic gene expression patterns in the co-culture vs non co-cultured pellets. ACCs only, BMSCs only, or BMSCs+ACCs (1:1) were differentiated in chondrogenic induction medium for 3 weeks and mRNA expression levels quantified by RT-qPCR. There was significantly higher gene expression of the chondrogenic markers COL2, ACAN, and SOX9 and the anti-angiogenic marker ChM-1 in the BMSC+ACC co-culture groups compared with the monoculture groups. By contrast, the gene expression of hypertrophic markers COL10, RUNX2, MMP-13, IHH, and ALPL and the angiogenic factor VEGF were lower in the mixed ACCs+BMSCs groups compared with the BMSCs alone group. The results are representative of experiments using three different patient samples with three technical replicates. Data are reported as means±s.d.; *P<0.05.
