Oral tolerance model. (a) Mice were fed five consecutive doses of antigen, followed 3 days later (day 8) by immunization with wOVA or peptide emulsified in CFA. On day 20, serum and fecal pellets were collected for antibody analysis, and the mice were killed; dLNs were isolated and subjected to in vitro restimulation with wOVA. Delayed-type hypersensitivity model. (b) Mice were fed five consecutive doses of antigen, followed 3 days later (day 8) by immunization with wOVA or peptide emulsified in CFA. On day 20, the mice were injected ID with OVA in the right ear and PBS in the left ear. Ear thickness was measured both 24 and 48 h after injection. On day 22, the mice were killed; the ears were collected and fixed in formalin for histological analysis, and dLNs were isolated and subjected to in vitro restimulation with wOVA. Allergic lung inflammation model. (c) Mice were fed five consecutive doses of antigen, followed 3 days later (day 8) by sensitization with wOVA in alum. Beginning on day 22, the mice received five consecutive daily intranasal doses of OVA in saline. Twenty-four hours after the final dose, serum and fecal pellets were collected for antibody analysis, and the mice were killed. BAL washes were performed, and lung tissue was obtained for histological analysis.
