Interleukin-21 (IL-21) depletion increases interferon (IFN)-γ and IL-17 production by lung CD4 T cells. Mice were immunized and challenged as described in Figure 2. Five days post challenge, lungs were harvested. CD4 T cells were sorted by magnetic activation cell sorter and stimulated overnight with (b) media or (c) αCD3/28 beads (10 μl per 106 cells). (a) CD4 T cells were stained for IFN-γ and IL-17 using specific catch and detection reagents (10 μl per 106 cells).
