Interleukin-21 (IL-21) depletion increases interferon (IFN)-γ and IL-17 production by lung CD4 T cells. Mice were immunized and challenged as described in Figure 2. Five days post challenge, lungs were harvested. CD4 T cells were sorted by magnetic activation cell sorter and stimulated overnight with (b) media or (c) αCD3/28 beads (10 μl per 106 cells). (a) CD4 T cells were stained for IFN-γ and IL-17 using specific catch and detection reagents (10 μl per 106 cells). The percentage of CD4 T cells secreting each cytokine was determined by flow cytometry and is shown in each dotplot. (d) Grouped data are also shown. The graphs are representative of two independent experiments of five mice per group. Student’s t-test result; ***P<0.001. TCR, T-cell receptor.
