Interleukin (IL)-1β-elicited signaling is dependent on mitogen-activated protein kinase (MAPK) activation. Rat late distal colon was mounted in Ussing chambers. Signaling inhibitors of mitogen-activated protein kinase/ERK kinase 1/2 (U0126, 10 μM) and p38 MAPK (SB202190, 10 μM) were added for 1 h to both chamber sides followed by addition of rat IL-1β (10 ng ml−1, 10 U ng−1) to the serosal chamber side for 6 h. Electrogenic sodium transport was measured as monovalent cation flux (JNa; μmol h−1 cm−2) after induction of epithelial sodium channel (ENaC) by aldosterone (3 × 10−9 M, 8 h) and consecutive inhibition by mucosally added amiloride.
