TL1A is a costimulatory cytokine for group 2 innate lymphoid cells (ILC2). (a) Surface expression of DR3 was measured by flow cytometry in Lin− Thy1+ lymphocytes vs. CD4+ T cells in mesenteric lymph nodes (mLN) and the intestinal lamina propria. (b) Lineage-negative/ICOS-positive cells were purified from interleukin-25 (IL-25)-treated Il13+/eGFP mice and treated with the indicated cytokines (IL-7 at 10 ng ml−1, IL-33 at 10 ng ml−1 and the indicated concentration of TL1A) for 72 h and then assayed for expression of the IL-13eGFP reporter by flow cytometry. Dot plots are gated on lineage-negative cells. (c) Mean fluorescence intensity (MFI) from data presented in (b). (d) Lineage (CD3CD4CD8CD19CD11bCD11cFcεR1Gr1)-negative ICOS-positive ILCs were purified from IL-25-treated Il13+/eGFP mice, and treated with 10 ng ml−1 of the indicated cytokines for 72 h before collection of supernatants for cytokine measurements.
