IL-36 can directly suppress the generation of Th17 responses while enhancing Th1 responses in vitro. (a) Magnetic activated cell sorting (MACS)-purified CD4+ T cells from C57BL/6 mice stimulated in vitro under neutral (Th0) or Th1-inducing conditions (IL-12 and anti-IL-4R) in the presence and absence of rIL-36α (200 ng ml−1) and analyzed for intracellular expression of IFNγ by fluorescence activated cell sorting (FACS). (b) MACS-purified CD4+ T cells from C57BL/6 il17a-egfp reporter mice stimulated in vitro under neutral (Th0) or Th17-inducing conditions (TGFβ, IL-6, anti-IL-4R, and anti-IFNγ) in the presence and absence of rIL-36α (200 ng ml−1) and analyzed for intracellular expression of green fluorescent protein by FACS. Data shown is representative of three independent experiments with similar results.
