Figure 1

IL-27 deficiency is associated with increased eosinophil and ILC2 frequencies in protease allergen induced pulmonary inflammation. (a) IL-27 concentrations in control (PBS) or papain-challenged mice were determined by ELISA. (b) Total RNA was isolated from lung tissue of PBS or papain-treated mice and the expression of IL-27 subunits (Ebi3, Il27p28) was analyzed by qRT-PCR relative to hprt. (c, d) Flow cytometric analysis of eosinophil (SSChighautofluorescence loSiglecF+CD11bhigh) and ILC2 (Lin-Thy1.2+KLRG1+) frequencies in lungs of papain-challenged wildtype, Ebi3−/− and Il27p28−/− mice. Graphs on the right depict the quantification of absolute ILC2 numbers per organ. (e) Intracellular IL-13 expression was quantified by flow cytometry after 3 h stimulation with PMA and Ionomycin. (f) IL-5 and IL-13 concentrations in BAL of papain-treated mice were determined by ELISA. Data represent 3 independent experiments with 4-5 mice per group. Error bars indicate s.e.m.
