  1. When calculating splice-site density, a pseudocount of one was added to the number of splice sites in an mRNA. For the comparisons between poly(A)-tail length and expression level, mRNA abundances were measured by RNA-seq; data for HeLa were from ref. 35. For the relationship between poly(A)-tail length and mRNA nuclear-to-cytoplasmic abundance ratio, measurements of nuclear and cytoplasmic mRNA abundance in HeLa cells were from ref. 47. mRNA half-lives for S. cerevisiae, HeLa and 3T3 mRNAs were from refs 48, 49, 50, 51, 52, 53, 54, 55, ref. 56 and ref. 31, respectively.
