Fluorescence loss is greatly reduced for the TMR labeled peptide, (fmoc)-K*A, under constant laser illumination in nitrogen purged 1 mM Trolox in methanol (top, green, curve, n=2802) versus imaging in methanol alone (bottom, red, curve, n=5942), imaged with consecutive 0.5 sec frames. Solid lines represent fits of each data set to a single exponential. Imaging in methanol/Trolox resulted in a t1/2 of approx. 105 seconds. * indicates TMR conjugated to lysine.
