(a) A 3D cytoskeletal network (3 × 3 × 3 μm) with an actin turnover rate of 30 s−1 with 25 μM actin, 5% ACPs (to actin) and 5% motors (to actin) at different time points. Motors start walking at t=0 s, and at t=3 s, the network is homogeneous. At t=20 s and 47 s, the network is progressively aggregating due to internal stress. (b) A network that has an actin turnover rate of 300 s−1 with 25 μM actin, 5% ACPs and 5% motors stays homogeneous and can sustain the motor-generated internal stress. Similar morphological states can be observed in 2D cortical networks (Supplementary Fig. 1). Teal, yellow and red are actin filaments, ACPs and motors, respectively.
