Socs1+/+ (5×105 cells per ml; black bars) and Socs1−/− (5×105 cells per ml; grey bars) BMDCs were stimulated with LPS (10 ng ml−1) and graded concentrations of 8-Br-cAMP (0, 33 and 100 mM) for 12 h with or without recombinant murine IL-6 (20 ng ml−1), IL-27 (100 ng ml−1), IL-4 (20 ng ml−1) and IL-13 (20 ng ml−1). (a) The levels of IL-12p70 and TNFα in the culture supernatant were measured by ELISA. Error bars indicate +s.d. Cells were stimulated in triplicate wells for each condition and s.d. was calculated from the values determined by ELISA. (b) The normalized antagonistic effect (% antagonism) of the indicated cytokines against the suppressive effect of cAMP for the production of IL-12p70.
