(a,b) Kinetic profiles of GIF:GM-CSF and GIF:IL-2 interactions characterized by BLI. In each case, biotinylated GIF was coupled to the surface of streptavidin coated BLI sensors, followed by binding measurements in different concentrations of GM-CSF/IL-2. Binding data was fitted using a 1:1 interaction model. (c,d) BLI measurements of the binding affinity of GM-CSF and IL-2 after saturating GIF bound sensors with IL-2 or GM-CSF respectively. After saturation with GM-CSF/IL-2, binding to the other cytokine is completely abolished (KD: NA=not applicable). (e,f) BLI measurements demonstrate that GIF does not bind to human orthologues of GM-CSF and IL-2. Displayed KD, kd and ka values represent the average of three replicate experiments.
