Upon externally applied stress, embedded beads undergo compression as shown in b,c. Bright-field images (b) serve to the contour of spheroids at equatorial plane. (c) Fluorescence of beads have been used to determine the surface before and after applied stress. (d) Present masks created upon IsoData threshold. The difference of surface area (magenta represents state before and cyan represents state after compression) as shown by superimposed images is converted into the strain, |V(P)−V0|/V0, for each bead found in focus.
