In case of non-farnesylated PEX19 CTD, the helix α1 is flexible in solution and only loosely attached to the helical core domain of the PEX19 CTD, while the lid adopts an open conformation. Farnesylation induces a specific, rigid arrangement of these two structural elements, thus forming a hydrophobic cavity, which then forms a high-affinity binding site for PMPs.
