Mouse adrenal chromaffin cells were stimulated with 70 mM K+ solution for 1 min (dotted line below trace), and catecholamine release measured from single cells using carbon fibre amperometry (a, upper trace). Individual current spikes indicate exocytosis from a single vesicle. Pre-incubation of cells with IC87114 (1 μM) for 2 min before stimulation caused a shift in release probability so that more vesicles were released earlier (a, bottom trace). This is quantified in (b) and values expressed as mean±s.e.m. (n=6 cells). (c) Catecholamine release measured using carbon fibre amperometry from a control cell (c, upper trace) and a cell treated with the combination of VO-OHpic (100 nM) and IC87114 (c, bottom trace). Normal secretion was observed in both cases and the treatment did not promote a shift in vesicle release probability compared with earlier time points (d). Data are expressed as mean±s.e.m. (n=8 cells).
