(a) Structure of the S3 region of the super-enhancer. Site S3 is characterized by STAT5 binding, extensive H3K27ac, Pol II binding (GSE74826) and the presence of eRNAs (based on total RNA-seq data). Transcripts coincided with site S3 and additional STAT5-binding sites. An eRNA associated with S3 was identified and characterized using 5′ and 3′ RACE (Supplementary Note 2). (b) No statistically significant differences in S3 eRNA levels between mammary tissue from wild type and the mutants were observed. One-way ANOVA with multiple comparison and alpha 0.01 was applied (P=0.26). Results are shown as the means±s.e.m. of independent biological replicates; ΔC n=9 (−/−); ΔD n=5 (−/−); ΔE n=6 (−/−); ΔCDE n=3 (−/−).
