Short-term T-cell lines were generated from peripheral blood stimulated with the indicated peptides. Viable CD3+CD4+ cells that bound the HLA-DR1/HA305−320 tetramer were sorted by flow cytometry, and a molecular analysis of expressed TRB gene rearrangements was conducted as described in the Methods. (a) Gating strategy used to identify viable CD3+CD4+ tetramer-positive events in: (b) line Flu1, (c) line Flu2 and (d) line Flu3.
