(a–c) In situ hybridization for Edn1 in WT hearts. At E12.5 (a), Edn1 expression is detected in the conotruncal cushion endocardium (arrowheads). At E13.5 (b) and E14.5 (c), the endothelium of proximal coronary arteries in the conal region expresses Edn1 (arrowheads). (d–l) Co-immunostaining for enhanced green fluorescent protein (EGFP) and β-galactosidase (β-gal) in the hearts of Wnt1-Cre/R26R; EdnraEGFP/+ mice. EGFP expression is driven by the endogenous Ednra promoter and reflects Ednra expression. At E12.5, EGFP signals were detected broadly in cardiomyocytes and aortic SMCs, and also in Wnt1-Cre-labelled cells in the condensed mesenchyme (arrow) and close to the endocardium (arrowhead; d–f). At E13.5 and E14.5, EGFP was expressed in Wnt1-Cre-labelled cells in the condensed mesenchyme (arrow) around coronary arteries (arrowheads; g–i,j–l).
