(a) Immunoblotting for Hsf-1, Hsp72, Hsp105, Hsp40 and Hsp90 in wild-type, Hsf-1-knockout wild-type, AR-97Q and Hsf-1-knockout AR-97Q mice (13 weeks old). (b–e) Quantitative analysis using densitometry revealed that the expression levels of Hsp72 in the spinal cord (b) and liver (c) were downregulated in heterozygous Hsf-1-knockout AR-97Q mice compared with AR-97Q mice (13 weeks old). No significant alterations in the signal intensity of the Hsp-immunoreactive bands were observed in skeletal muscle (d) or testis (e). Data are shown as the ratio of the intensity of each molecule to Gapdh. (f–h) Quantification of Hsp70A1 mRNA levels using RT-PCR in the spinal cord (f), liver (g) and skeletal muscle (h). *P<0.05 by unpaired t-test (n=7) (b–e), and (n=3) (f–h).
