To characterize the supernumerary centriolar structures that are observed following LGALS3BP depletion, the presence of markers for daughter centrioles (CNTROB) and parental centrioles (glutamylated tubulin) on those structures in comparison with centrin was quantified. (a) Immunofluorescence microscopy analysis of U2OS cells treated with siRNA against LGALS3BP revealed that the number of both parental and daughter centrioles stays constant, whereas the number of centrin-positive structures increases. Magnifications of the centrioles are shown in the inserts on the bottom of each image. Nuclei are labelled with DAPI. Scale bars, 5 μm.
