(a) Loss of Dnmt1 focal staining pattern in cells replicating in mid or late S phase in Uhrf1−/− ES cells. Both E14 and Uhrf1−/− ES cells were immunostained for Dnmt1 and costained by DAPI. The cells with Dnmt1 focal staining pattern were observed only for the wild-type E14 cells. These cells were in mid or late S phase according to EdU labelling (Supplementary Fig. S7 and references). Scale bar, 10 μm. (b) The immunostaining analysis for rescue of Dnmt1 focal staining pattern in each stable cell line was performed with a mixture of each type of stable cells and GFP-Uhrf1−/− ES cells in ~1:1 ratio. The representative merged images of GFP and DNMT1 show that the presence of Dnmt1 focal staining cells (marked by arrows) in comparison with the GFP-Uhrf1−/− ES cells except for the double mutant stable cells (YP191/192AA+YT478/479AA).
