MEFs from Pellino3+/+ and Pellino3−/− embryos were co-transfected with a NFκB-regulated firefly luciferase gene and an expression vector encoding a super repressor form of IκBα (IκBαSR) or an empty vector (EV). Cells were treated in the absence (Control) or presence of murine TNF (50 ng ml−1) and cycloheximide (CHX) (10 μg ml−1) for 8 h. (a) Data are presented as normalized luciferase expression relative to unstimulated cells. (b) Cells were analysed by flow cytometry for percentage of cells staining positive for annexin V. Data represent the mean +/− s.e.m. of two independent experiments. (c) Cell lysates were probed by immunoblotting for expression levels of PARP, IκBα and β-actin. A cleaved form of PARP is indicated by an arrow. Full-length images of immunoblots are shown in Supplementary Fig. S16.
