(a) Western blots illustrate the effects of H2O2 in GFP-, GFP-TDP-43-FL- or GFP-TDP-43-Q303P-expressing cells. Treating transfected cells with 0.2 mM H2O2 for 30 min, followed by a 24-h recovery period, significantly increased p-AKT and PARP levels and decreased pro-caspase 3 levels in GFP- and TDP-43-FL-expressing cells but not in GFP-TDP-43-Q303P-expressing cells. (b) After H2O2 treatment, AKT, SIRT1 and HDAC6 expression was downregulated in GFP-TDP-43-FL- and GFP-TDP-43-Q303P-expressing cells but not in GFP-expressing cells. (c) Treatment with 0.2 mM H2O2 increased PGC-1α expression in TDP-43-FL-expressing cells. (d) Treatment with 0.2 mM H2O2 decreased HSP27 expression in all the transfected cells.
