Figure 2: The JNK pathway is activated in netA-IR discs and does not regulate NetA.

(a–f) JNK pathway activity assessed by expression of two JNK targets, puc-lacZ (anti-β-gal, a – c) and anti-MMP1 (d–f). lacZ is expressed in Ubx-GAL4, puc-lacZ control disc and Ubx>netA-IR, puc-lacZ disc (b) but is lost in Ubx>bskDN, puc-lacZ discs (c) (although some lacZ expression persists outside the Ubx-GAL4 expression domain—arrowheads). Punctate anti-MMP1 staining was observed in the stalk region of control Ubx-GAL4/+ (d), and Ubx>netA-IR (e) third instar discs, but not in Ubx>bskDN discs (f). (g–i) Control Ubx-GAL4, puc-lacZ/+ disc showing expression of NetA puncta (h) and puc-lacZ in peripodial cells (i, arrowheads). (j–l) In hepR75/Y;Ubx-GAL4,puc-lacZ/+ discs, puc-lacZ expression is lost (l), but NetA expression appears normal (k). Scale bars, 50 μm (a–c,g–l) and 10 μm (d–f).
