Figure 5

(a) W/T or Itgam−/− BM-ΜΦs and BM-DCs were treated with the indicated doses of LPS and the amounts of RANTES secreted were measured using ELISA at 6 h and O/N. Data are mean±s.e.m. from three independent experiments with three mice per group. (b) W/T or Itgam−/− BM-ΜΦs and BM-DCs were left untreated (CM) or treated with LPS (1 μg ml−1) for the time indicated. The presence of phosphorylated (p-)TBK-1 and IRF3 was assessed using western blot. Quantification of band intensity showing the fold increase in phospho-protein levels relative to untreated cells and normalized to total protein levels. Data are mean±s.e.m. from three independent experiments. (c) W/T or Itgam−/− BM-DCs were left untreated (CM) or treated with 1 μg ml−1 of LPS for 24 h. DC maturation markers (MHC class II, CD80 and CD86) were assessed using flow cytometry. Data are expressed as delta MFI (MFI=test-isotype control). Data are representative of three independent experiments (mean±s.e.m., n=3) *P<0.05, **P<0.01, ***P<0.005 (t-test).
