(ac) Female W/T or Itgam−/− mice (n=3) were given i.n. twice 100 μg HYDby peptide or peptide plus 3 μg LPS. Splenic CD11c+ cells isolated 24 h later were: (a) immediately stimulated with PMA/ionomycin for 6 h and the % of IL-6+ cells was determined using flow cytometry. The amount of secreted RANTES at 24 h was measured using ELISA; (b) cocultured with Marilyn T cells and the T-cell proliferation was assessed by 3H-thymidine uptake at 48 h. (c) Percentage of CD11c+ IL-6-producing cells in the 3-day DC/T cocultures was quantified by intracellular staining. Data are shown as mean±s.e.m., n=3, *P<0.05, **P<0.01 (t-test).
