Figure 1: Sema3A enhances immunostaining of GluA2/3 and GRIP1 in dendrites.

From: Plexin-A4-dependent retrograde semaphorin 3A signalling regulates the dendritic localization of GluA2-containing AMPA receptors

Figure 1

(a) Sema3A enhanced immunostaining of GluA2, GluA3 and GRIP1 without modifying those of GluA1 and GluN1 in the MAP2-positive dendrites of cultured hippocampal neurons. Sema3A also enhanced expression levels of GluA2 in the cell surface of dendrites. (b) Sema3A applied at the axonal region, but not somatodendritic region (S+D), enhanced immunostaining of GluA2 in the dendrites of cultured hippocampal neurons. Representative images of the somatodendritic regions (indicated in the box in Supplementary Fig. 1e) are shown. Arrowheads in b indicate axon. Normalized mean immunostaining intensity of dendrite per neuron is shown in c (n=48–54 neurons from more than three independent cultures). Magnified images of boxed areas in upper panels are shown in lower panels (a and b). Scale bars in a, 50 μm (upper panel); 10 μm (lower panel); b, 20 μm (upper panel) or 10 μm (lower panel). *P<0.01 by ANOVA compared with vehicle control. Data are presented as mean±s.e.m.
