Anti-Flag M2 affinity gel was used for immunoprecipitation (IP); antibodies to CaM or Flag were used for western blot (WB). (b) The IQ motif is required for the interaction of IQCG with CaM. ΔIQCG refers to the IQCG construction with IQ motif deleted. (c) SPR-binding experiments of human IQ motif over immobilized CaM in Ca2+-free (left panel) or Ca2+-supplemented condition (right panel). The concentrations of IQCG IQ motif were 8.4–50 nM and 20–320 nM, respectively. The curves represent direct fits of the 1:1 Langmuir-binding model to the data.
