E13, E14 and E15 DRGs were grown on cell culture inserts. NT3 (a,b,d) or NGF- (c) were added to the culture media to promote growth of proprioceptive or cutaneous axons, respectively. Cell bodies (a) and axons (bd) were extracted separately from the two sides of the insert membrane and analysed by western blot with antibodies against Nrp1. (d) E14 and E15 cultures were supplemented with the MP inhibitor TAPI-1 or DMSO (carrier). Immunoblots for tubulin βIII (neuronal specific) and actin were used as loading controls. Molecular weight markers (kDa) are indicated on the right. Full-scanned western blots are presented in Supplementary Fig. 6.
