Exponentially growing C2C12 myoblasts were transfected with the HuR or control (Ctr) siRNAs. (a) RNA was prepared 48 h after transfection with HuR or Ctr siRNAs. Northern blot was performed using radiolabelled probes against NPM, MyoD, HuR mRNAs and 18S (loading control). The band intensities of NPM, MyoD mRNAs and 18S were determined using ImageQuant Software. The NPM and MyoD mRNA levels were normalized on 18S rRNA level. (b) Forty-eight hours after transfection with HuR or Ctr siRNAs, whole-cell extracts were prepared and western blot was performed using antibodies against NPM, HuR, myogenin and α-tubulin (loading control). ImageQuant was used to determine the NPM level, normalized on α-tubulin level. In the histograms, the siRNA HuR condition was plotted relative to the siRNA Ctr condition±s.e.m. of three independent experiments *P<0.01, **P<0.001 (t-test).
